Targeted Liquid Biopsy Assay

OncoSELECT® — targeted liquid biopsy from plasma

A kit developed specifically for liquid biopsy. 74 key genes, LOD down to 0.25% VAF for SNVs and indels. From a standard 20 ng of cfDNA extracted from plasma, you obtain the full range of variants (SNVs, indels, CNVs, fusions, TERT promoter alterations and splice variants) with a clinical report within 5 days.

  • 74 Genes in the panel
  • 0.25% LOD for SNVs and indels
  • 5 days From sample to clinical report
  • > 99% Specificity across variants
About the test

Liquid biopsy for patients with solid tumours

OncoSELECT® is a hybrid-capture panel for tracking 74 key cancer-relevant genes in circulating tumour DNA (ctDNA). It was developed and optimised for liquid biopsy from blood plasma.

The solution also includes the OncoKDM® platform for complete data analysis through to a clinical report, including recommended treatment and clinical trials.

Specifications

What OncoSELECT® offers

74 genes

Hybrid-capture panel covering key cancer-relevant genes across the most common solid tumours.

Broad variant spectrum

SNVs, indels, CNVs, gene fusions / translocations, TERT promoter alterations and splice variants in a single panel.

20 ng cfDNA

The recommended input is 20 ng of high-quality cfDNA from plasma. Compatible with standard extraction kits (Norgen, QIAGEN, etc.).

~ 20,000× coverage

120 M paired-end reads per sample provide an average depth of 19,000–20,000×, sufficient to detect low-frequency variants.

UMI

OncoSELECT uses UMI (Unique Molecular Identifier) technology to increase the accuracy of low-frequency variant detection.

Sequencing platforms

Validated on NextSeq 550 High Output and NovaSeq 6000 (SP / S1 / S2). Validation on Element and MGI is ongoing.

Analytical performance

Verified parameters across variant types

The limit of detection (LOD) is defined as the lowest VAF at which at least 95% of replicates yield a positive result. Data from the OncoDNA validation study:

Variant type LOD (VAF) Sensitivity Specificity Accuracy
SNVs 0.25% 96.88% > 99% > 99%
Indels 0.25% 96.93% > 99% > 99%
Fusions 0.5% 97.60% > 99% > 99%
CNVs 1% > 99% > 99% > 99%
Example number of samples per run
  • NextSeq 550 High Output 6 samples / run
  • NovaSeq 6000 SP 12 samples / run
  • NovaSeq 6000 S1 24 samples / run
  • NovaSeq 6000 S2 48 samples / run
Workflow

From sample to report in 5 days

All preparation takes place in your own laboratory. OncoDNA supplies the kit, the analytical processing and a clinically oriented report in the OncoKDM® platform, with expert review before delivery.

  1. 1

    cfDNA extraction

Extraction of circulating DNA from patient plasma. The recommended input for library preparation is 20 ng of high-quality cfDNA.

  2. 2

    Library preparation

Sequencing library preparation, compatible with Twist-based protocols and automation on Opentrons Flex.

  3. 3

    Hybrid capture

Targeted enrichment of 74 genes using the Twist Bioscience hybrid-capture protocol with uniform coverage.

  4. 4

    Sequencing

Compatible with Illumina sequencers. Validation on Element and MGI is ongoing.

  5. 5

    OncoKDM® report

From secondary and tertiary analysis to a clinical report in OncoKDM®.

Clinical applications

Where OncoSELECT® delivers answers

Treatment selection

Identification of actionable variants that can be targeted with available therapy, directly from plasma and without the need for a tissue biopsy.

Disease heterogeneity

A view across all metastatic sites at once, which a single tissue biopsy cannot capture.

Clinical trial eligibility

Screening patients for clinical trials based on specific variants.
