Hedera Profiling RNA — a 43-gene panel for fusions and expression

Hedera Profiling RNA complements the DNA panels where you need to work with the transcript. It detects gene fusions independently of the fusion partner, as well as splice variants and clinically significant exon skipping. Alongside rearrangements, it also profiles gene expression.

About the test

An RNA panel for fusions and splice variants

Some clinically significant rearrangements are hard to find in DNA. The breakpoint lies in a long intron, or the fusion partner is unknown. Hedera Profiling RNA approaches them from the other side: it works with the transcript, where a fusion appears as two exons joined together.

Probes are placed at exon boundaries across all coding exons of the selected genes. The panel therefore detects a fusion regardless of the fusion partner and distinguishes individual isoforms. Alongside fusions, it reads the expression profile and splice variants, including exon skipping.

Specifications

What the HP RNA panel covers in one test

43 genes

Genes with clinically significant rearrangements and expression markers.

Fusions without a known partner

Probes at exon boundaries detect a fusion regardless of what the gene is fused with.

Splice variants

The panel distinguishes isoforms and detects clinically significant exon skipping.

Expression profiling

Alongside rearrangements, it also reads the expression profile of the genes covered.

Twist Bioscience technology

Hybrid capture on standard laboratory equipment, with nothing extra to buy.

Hedera Prime software

Data analysis and reporting in one place, from secondary analysis to the clinical report.

Panel content

The complete list of genes in the panel

Based on the original Hedera Dx specification. Genes shown in dark type have probes at exon boundaries across all coding exons. For these genes, the panel detects a fusion independently of the fusion partner.

Workflow

From sample to clinical report

The entire workflow takes place in your laboratory. Hedera Dx supplies the kit, the protocol and the Hedera Prime software. The output is a clinical report for the oncologist or molecular pathologist.

  1. 1

    RNA extraction

Total RNA is extracted from the sample.

  2. 2

    cDNA synthesis and library preparation

Twist Bioscience technology. Runs on standard laboratory equipment, with nothing extra to buy.

  3. 3

    Hybrid capture

Hybrid capture of 43 genes with probes targeted at exon boundaries.

  4. 4

    Sequencing

The panel runs on standard Illumina and Element Biosciences sequencers.

Implementation support

Hedera Comply — tailored implementation under IVDR

The panel comes with the Hedera Comply programme. It takes your laboratory all the way from installation to operation under IVDR.

  • Hands-on training on site in your laboratory
  • Laboratory procedures and software training
  • Review of the first runs and validation
  • Ready-made technical documentation for IVDR
  • Ongoing support, including after implementation
