PacBio: HiFi long‑read sequencing

PacBio (Pacific Biosciences) is a US manufacturer of sequencing platforms focused on high-accuracy long-read technology. Its HiFi sequencing combines the best of the short-read and long-read worlds: long reads, high accuracy and a comprehensive view of the genome.

Product family

Two systems, one HiFi family

PacBio offers two platforms built on the same HiFi chemistry, for laboratories and projects of different sizes.

High-throughput

Revio

Benchtop

Vega

Compact · Benchtop sequencer

A benchtop long-read sequencer that brings the same HiFi accuracy to smaller laboratories. A benchtop format, a simple workflow and a lower initial investment, with no compromise on data quality.

  • Compact benchtop design
  • The same HiFi technology as Revio
  • Ideal for targeted panels and smaller projects
  • Quick start-up and easy operation

What PacBio focuses on

From research to diagnostics

PacBio builds technologies for scientists and clinicians who need a complete view of the genome, from individual bases through structural variants to methylation.

Human genetics

Whole-genome and targeted panel sequencing for clinical research, rare-disease diagnostics and population studies.

Plants and animals

Plant and animal research using accurate long-read sequencing delivers actionable genomic insights for addressing global challenges.

Microbiome

Uncover the complex composition of microbial communities and gain deeper insight into metagenomics with accurate long-read sequencing.

Cancer genomics

Enables comprehensive analysis of tumour genomes, epigenomes and transcriptomes with a single technology.

How HiFi works

Three pillars of highly accurate HiFi reads

In each run, PacBio circularises the DNA and reads the same fragment repeatedly. The resulting consensus eliminates random errors and produces highly accurate long-read data.

SMRT Cells

HiFi sequencing takes place on unique SMRT Cells (Single Molecule, Real-Time), in wells called ZMWs (Zero-Mode Waveguides). The polymerase reads the native DNA molecule in real time: no PCR, no amplification bias.

Repeated reading of a circular molecule

The SMRTbell library joins the ends of the molecule into a circle. The polymerase then travels around this circular DNA repeatedly, so a single molecule yields 8–15 independent reads.

Consensus HiFi read

A consensus is calculated from the repeated passes: a single, highly accurate long read with a Q33+ score. Methylation information from the native DNA is preserved at the same time.

Key benefits of HiFi technology

One technology. Comprehensive answers.

Long reads (up to 25 kb)

A single read spans entire genes, regulatory regions and structural variants that are beyond the reach of short-read technologies.

Q33 accuracy (99.95%)

Circular consensus sequencing reads the same molecule repeatedly, resulting in short-read-level accuracy.

Methylation detection in a single run

Epigenetic information directly from native DNA, with no need for chemical conversion or a separate library.

Detection of all variant types

SNVs, indels, structural variants, repetitive regions and phasing, all from a single experiment.

De novo assembly

Accurate de novo assembly of genomes without a reference sequence.

Reproducibility

Consistent results across instruments and runs, which is critical for clinical and population applications.

New in 2026

SPRQ-Nx — the new sequencing chemistry!

SPRQ-Nx is the latest generation of PacBio sequencing chemistry, taking HiFi technology even further. It delivers a higher data yield from each SMRT Cell and better accuracy, while remaining fully compatible with existing workflows.

With the new SPRQ-Nx chemistry, SMRT Cells can be reused, reducing sequencing costs by up to 30%.
